Application
PIK93 has been used to examine if the accumulation of SNX27–retromer cargoes in FAM21 (family with sequence similarity 21) depleted cells is caused by an elevation of phosphatidylinositol 4-phosphate [PI(4)P] levels at the Golgi.
Biochem/physiol Actions
PIK-93 inhibits of replication of enterovirus including polio and hepatitis C. PIK93 is a potent, selective, ATP competitive and reversible inhibitor of PI3Kγ and PI4KIIIβ (IC50 = 16 nM and 19 nM, respectively). Also PIK93 moderately inhibits other members of the family including p110δ, PI 3-KC2β, hsVPS34, ATM, p110β and PI 4-KIIIα (IC50 = 0.120, 0.140, 0.320, 0.490, 0.590 and 1.1 µM, respectively). PIK93 blocks ceramide transfer protein-mediated ceramide traffic between endoplasmic reticulum and Golgi.
PIK93 is also known as N-(5-(4-Chloro-3-(((2-hydroxyethyl)amino)sulfonyl)phenyl)-4-methyl-2-thiazolyl)-acetamide. It inhibits poliovirus (PV) and coxsackievirus B3 (CVB3) viral RNA production and virus replication. PIK93 also prevents translation mediated by internal ribosome entry site (IRES).
