Storage and Stability

Caspase-1 (UniProt: P29466; also known as EC:3.4.22.36; CASP-1, Interleukin-1 beta convertase, IL-1BC, Interleukin-1 beta-converting enzyme, ICE, IL-1 beta-converting enzyme, p45) is encoded by the CASP1 (also known as IL1BC, IL1BCE) gene (Gene ID: 834) in human. Caspases are secreted as latent single chain zymogens, which are further processed to create active caspases. The zymogen form of caspase-1 has an N-terminal propeptide (aa 1-119) and another propeptide (aa 298-316), which are cleaved off to generate p20 (aa 120-297) and p10 (aa 317-404) subunits. Its CARD domain is localized in amino acids 1-91. The two subunits (p20 an p10) heterotetramerize to form two anti-parallel arranged heterodimers. Caspase-1 is a thiol protease that cleaves IL-1 beta between an Asp and an Ala, releasing the mature cytokine that is involved in a variety of inflammatory processes. It is a component of the inflammasome protein complex. It has a preferred cleavage site of Tyr-Val-Ala-Asp (YVAD).
