Application

Anti-Angiopoietin-1, clone 2F9 ZooMAb, Cat. No. ZRB1522, is a recombinant Rabbit monoclonal antibody that specifically targets Angiopoietin-1 and is tested for use in Affinity Binding Assay, Immunocytochemistry, and Immunohistochemistry (Paraffin).

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Angiopoietin-1 in HeLa, A431, HepG2, HUVEC, and NIH 3T3 cells.Affinity Binding Assay: A representative lot of this antibody bound Angiopoietin-1 peptide with a KD of 7.2 x 10-7 in an affinity binding assay.Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

General description

ZooMAb antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. ZooMAb antibodies are reliably available and ready to ship when you need them.Learn more about ZooMAb here.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal region of human Angiopoietin-1.

Legal Information

ZooMAb is a registered trademark of Merck KGaA, Darmstadt, Germany

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Specificity

Clone 2F9 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Angiopoietin-1. It targets an epitope within 22 amino acids from the N-terminal region.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Target description

Angiopoietin-1 (UniProt: Q15389; also known as ANG-1) is encoded by the ANGPT1 (also known as KIAA0003) gene (Gene ID: 284) in human. ANG-1 is a homooligomeric secreted glycoprotein that binds and activates TEK/Tie2 receptor by inducing its dimerization and tyrosine phosphorylation. It is synthesized with a signal peptide (aa 1-15), which is subsequently cleaved off to produce the mature form. Four forms of angiopoietins have been described (ANG1-ANG4) that serve as ligands for Tie2, a tyrosine kinase that is found on endothelial cells. ANG-1 serves as an activator of Tie2 and ANG-2 acts as an antagonist of Tie2 and prevents its activation. ANG-1 plays a role in the regulation of angiogenesis, endothelial cell survival, proliferation, migration, adhesion and cell spreading. It acts as a chemoattractant for endothelial cells and promotes endothelial cell sprouting and facilitates tissue invasion by nascent blood vessels through activation of MMPs. ANG-1 is also required for normal angiogenesis and heart development during embryogenesis. After birth, ANG-1 is shown to activate or inhibit angiogenesis, depending on the context. It inhibits angiogenesis and promotes vascular stability in quiescent vessels. Two isoforms of ANG-1 have been described that are produced by alternative splicing. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
