Reconstitution

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Target description

Hepatocyte nuclear factor 4-alpha (UniProt: P41235; also known as HNF-4-alpha, Nuclear receptor subfamily 2 group A member 1, Transcription factor 14, TCF-14, Transcription factor HNF-4) is encoded by the HNF4A (also known as HNF4, NR2A1, TCF14) gene (Gene ID: 3172) in human. HNF-4 is a transcriptional regulator that controls the expression of hepatic genes during the transition of endodermal cells to hepatic progenitor cells, facilitating the recruitment of RNA pol II to the promoters of target genes. It activates the transcription of CYP2C38 and represses the CLOCK-ARNTL/BMAL1 transcriptional activity. It is reported to be essential for circadian rhythm maintenance and period regulation in the liver and colon cells. It can undergo phosphorylation on tyrosine residues and that is shown to be important for its DNA-binding activity. However, phosphorylation at serine 313 by AMPK can reduce its ability to form homodimers and bind DNA. Mutations in HNF4A gene are known to cause type II diabetes. Changes in the HNF4A gene can reduce the amount of insulin that is produced by the pancreas.
