Anti-PIAS, clone 1O17 ZooMAb, Cat. No. ZRB1450, is a recombinant Rabbit monoclonal antibody that specifically targets PIAS and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PIAS in lysates from K562, L6, Sh-SY5Y, and NIH3T3 NE, and HeLa cell nuclear extracts. Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PIAS in SH-SY5Y cells.Affinity Binding Assay: A representative lot of this antibody bound recombinant E3 Sumo protein fragment with a KD of 4.7 x 10-8 in an affinity binding assay.Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected PIAS in human testis tissue sections.Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Target description

E3 SUMO-protein ligase PIAS1 (UniProt: O75925; also known as DEAD/H box-binding protein 1, E3 SUMO-protein transferase PIAS1, Gu-binding protein, GBP, Protein inhibitor of activated STAT protein 1, RNA helicase II-binding protein) is encoded by the PIAS1 (also known as DDXBP1) gene (Gene ID: 8554) in human. PIAS is an E3-type small ubiquitin-like modifier (SUMO) ligase that is expressed in numerous tissues with highest levels found in the testis. It stabilizes the interaction between UBE2I and the substrate and also acts as a SUMO-tethering factor. PIAS plays a crucial role as a transcriptional coregulation in various cellular pathways, including the STAT pathway, the p53 pathway and the steroid hormone signaling pathway. The effects of this transcriptional coregulation, transactivation or silencing, may vary depending upon the biological context. It is known to enhance the sumoylation of MTA1 and may also participate in its paralog-selective sumoylation. It also plays a role in adipogenesis by promoting the SUMOylation and degradation of CEBPB. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
