Application
Immunocytochemistry Analysis: A 1:100-1,000 dilution from a representative lot detected LMX-1 in Neuro2A cells. Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected LMX-1 in mouse embryo and human brain tissue sections.Affinity Binding Assay: A representative lot of this antibody bound recombinant Human LMX-1 protein with a KD of 1.1 x 10-6 in an affinity binding assay.Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

General description
ZooMAb antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. ZooMAb antibodies are reliably available and ready to ship when you need them.Learn more about ZooMAb here.
Immunogen
GST/His-tagged recombinant fragment correspond to 80 amino acids from the C-terminal region of Hamster LMX-1..
Legal Information
ZooMAb is a registered trademark of Merck KGaA, Darmstadt, Germany
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Specificity
Clone 4L12 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects LIM homeobox transcription factor 1-alpha (LMX-1). It targets an epitope within 80 amino acids from the C-terminal region.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description
LIM homeobox transcription factor 1-alpha (UniProt: Q04650; also known as LIM/homeobox protein LMX1A, LIM/homeobox protein 1, LMX-1, LMX1A) is encoded by the LMX1A (also known as LMX1) gene (Gene ID: 101839559) in hamster. LMX1A is a member of the group of LIM-homeobox-containing genes that encode LIM-homeodomain (LIM-HD) protein. LMX-1 is a transcription factor, is expressed in the dorsal mesenchyme and acts by binding to an A/T-rich sequence. It is induced within the ventral midbrain as a response to early signaling It is required for development of the roof plate and for specification of dorsal cell fates in the CNS and developing vertebrae. LMX-1 is reported to play a significant role in of mesencephalic dopamine neurons (mesDA) differentiation of human embryonic stem cells. It has high potential for stimulating the production mesDA that may have applications in the treatment Parkinson disease. Overexpression of LMX-1 is reported to inhibit tumor growth and metastasis. It is shown to be inactivated in gastric cancers. Its tumor growth inhibition is suggested to be due to its inhibitory effect on c-Myc expression. Serum levels of LMX-1 have been used as an effective biomarker for early detection of gastric cancers. Hamster LMX-1 contains two zinc binding domains (aa 33-92 and 93-154). Its DNA binding domain is localized to amino acids 195-254. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
