Target description
Cardiac phospholamban (UniProt: P26678; also known as PLB) is encoded by the PLN (also known as PLB) gene (Gene ID: 5350) in human. Phospholamban is a cardiac muscle-specific, 6 kDa, single-pass membrane protein that is shown to associate reversibly into pentamers. It modulates the contractility of the heart muscle in response to physiological stimuli via its effects on ATP2A2. It modulates calcium re-uptake during muscle relaxation and plays an important role in calcium homeostasis in the heart muscle. The degree of ATP2A2 inhibition depends on the oligomeric state of phospholamban. It is reported to tonically inhibit the SERCA pump by increasing its Km for Ca2+. Phosphorylation of phospholamban by protein kinase A at serine 16 is shown to relieve this inhibition, increasing the SR Ca-ATPase activity and lowering the cytoplasmic [Ca2+]. It can also undergo phosphorylation at threonine 17 by CaMK2 and in response to beta-adrenergic stimulation. Mutations in PLN gene have been linked to cardiomyopathies characterized by ventricular dilation and impaired systolic function that result in congestive heart failure and arrhythmia.
