No. ZRB1173, is a highly specific recombinant rabbit monoclonal antibody that detects JNK/SAPK phosphorylated on Thr183 and Tyr185 and has been tested for use in Immunocytochemistry, Immunohistochemistry, Peptide Inhibition Assay, and Western Blotting.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected phospho JNK (Thr183, Tyr185) in NIH3T3 cells treated with Anisomyocin.Immunohistochemistry (Paraffin) Analysis: A 1:100-1,000 dilution from a representative lot detected phospho JNK (Thr183, Tyr185) in human colon cancer and human ovarian cancer tissue sections. Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected phospho JNK (Thr183, Tyr185) in HeLa cells treated with Anisomycin treatment.Peptide Inhibition Analysis: A 1:1,000 dilution from a representative lot was used with NIH3T3 cells treated with Anisomyocin ((25 ug/ml for 30 min following overnight serum starvation) for peptide block analysis.Note: Actual optimal working dilutions must be determined by the end user as specimens and experimental conditions may vary.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Mitogen-activated protein kinase 8/9 (UniProt: P45983/P45984; also known as MAP kinase 8/9, MAPK8/9, Stress-activated protein kinase 1c/1a, SAPK1c/SAPK1a, Stress-activated protein kinase JNK1/ JNK2, c-Jun N-terminal kinase 1/2) is encoded by the MAPK8/9 (also known as JNK1/JNK2, PRKM8/9, SAPK1/2, SAPK1C/1A) gene (Gene ID: 5599/ 5601) in human. SAPK/JNK is a serine/threonine-protein kinase that is involved in various biological processes, including cell proliferation, differentiation, migration, transformation, and programmed cell death. It is activated in response to extracellular stimuli such as proinflammatory cytokines or physical stress. Both JNK1 and JNK2 contain a TXY motif (aa 183-185) that contains the threonine and tyrosine residues whose phosphorylation activates these kinases. Protein kinase domain of SAPK/JNK is localized to amino acids 26-321. Phosphorylation and activation of SAPK/JNK is achieved by two dual specificity kinases, MAP2K7 and MAP2K4. Following their activation SAPK/JNK phosphorylate several transcription factors, primarily components of AP-1 such as JUN, JDP2, and ATF2 and thus regulates AP-1 transcriptional activity. It promotes apoptotic cell death in stressed cells by phosphorylating key regulatory factors including p53/TP53 and Yes-associates protein YAP1. The immunogen sequence used to generate this clone is conserved in both JNK1 and JNK2. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
