General description

ZooMAb antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. ZooMAb antibodies are reliably available and ready to ship when you need them.Learn more about ZooMAb here.

Immunogen

KLH-conjugated Linear Peptide corresponding to the N-terminal of Human Androgen Receptor.

Legal Information

ZooMAb is a registered trademark of Merck KGaA, Darmstadt, Germany

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Specificity

Clone 3K11 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Androgen Receptor. It targets an epitope within the first 21 amino acids.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Target description

Androgen receptor (UniProt: P10275; also known as Dihydrotestosterone receptor, Nuclear receptor subfamily 3 group C member 4) is encoded by the AR (also known as DHTR, NR3C4) gene (Gene ID: 367) in human. Androgen receptor is a member of the family of NR3 ligand-dependent transcription factors that is composed of three domains: a modulating N-terminal domain, a DNA-binding domain and a C-terminal ligand-binding domain. In the presence of bound steroid, the ligand-binding domain interacts with the N-terminal modulating domain, and thereby activates AR transcription factor activity. It is predominantly cytoplasmic in its unlighted form, but translocates to the nucleus upon activation following ligand-binding and bind to DNA as homodimer. The androgen receptor associates with a number of adaptor-like proteins and functions as a complex that may induce or repress transcription depending on whether its associated proteins are co-activators or co-repressors that affect transcription possibly by modifying chromatin. Here co-activators are involved in histone acetylation and transcriptional activation whereas co-repressors are involved in histone deacetylation and transcriptional repression. The androgen receptor is considered to be important for determining sex and male characteristics. Defects in androgen receptor have been linked to metastatic prostate cancer. The mutated receptor stimulates prostate growth and metastases development despite of androgen ablation. Four different isoforms of androgen receptor have been described that are produced by alternative splicing. Isoform 3 and isoform 4 are reported to lack the C-terminal ligand-binding domain and therefore constitutively activate the transcription of a specific set of genes independently of steroid hormones. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
