Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description
Serine/threonine-protein phosphatase PGAM5, mitochondrial (UniProt: Q96HS1; also known as EC:3.1.3.16, Bcl-XL-binding protein v68, Phosphoglycerate mutase family member 5) is encoded by the pgam5 GENE (Gene ID: 192111) in human. PGAM5 is a single-pass mitochondrial membrane protein that displays phosphatase activity for serine/threonine residues and is shown to dephosphorylate and activate MAP3K5 kinase. It does not display any phosphoglycerate mutase activity. PGAM5 usually exists as a dimer, but can also form ternary complex with Nuclear Factor, Erythroid 2 Like 2 (NFE2L2) and Kelch Like ECH Associated Protein 1 (KEAP1). Its transmembrane domain is localized to amino acids 7 to 29. A 35 amino acid sequence at its N-terminal function as a non-cleaved mitochondrial targeting sequence that targets the protein to the cytosolic side of the outer mitochondrial membrane. Two isoforms of PGAM5 have been described that are produced by alternative splicing. Both isoforms are reported to be phosphorylated by RIPK1/RIPK3 complex under conditions of necrosis and this phosphorylation is shown to increase PGAM5 phosphatase activity. Overexpression of isoform 1 results in clustering of mitochondria around the nucleus whereas isoform 2 overexpression results in the formation of disconnected punctuate mitochondria distributed throughout the cytoplasm.
