Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Target description

T-cell surface glycoprotein CD8 alpha chain (UniProt: P01732; also known as T-lymphocyte differentiation antigen T8/Leu-2, CD8a) is encoded by the CD8A (also known as MAL) gene (Gene ID: 925) in human. CD8a is a single-pass type I integral membrane protein with one Ig-like V-type domain (aa 22-135) that plays an essential role in the immune response and serves multiple functions in responses against both external and internal offenses. CD8a is produced with a 21-amino acid signal peptide that is subsequently cleaved to produce the mature form. CD8 is generally present as a heterodimer of alpha and beta chains linked by two disulfide bonds, however, it can also form homodimers. The CD8 alpha/alpha homodimer is expressed on a subset of gamma/delta TCR-bearing T cells, NK cells, intestinal intraepithelial lymphocytes, and lymphoid dendritic cells. CD8a functions primarily as a co-receptor for MHC class I molecule:peptide complex. CD8a is reported to interact simultaneously with the T-cell receptor (TCR) and the MHC class I proteins presented by antigen presenting cells. In NK-cells, the presence of CD8a homodimers at the cell surface provides a survival mechanism allowing conjugation and lysis of multiple target cells. CD8a homodimer molecules also promote the survival and differentiation of activated lymphocytes into memory CD8 T-cells. Cytoplasmic domains of CD8 provide binding sites for the tyrosine kinase lck, facilitating intracellular signaling events that lead to T cell activation, development, and cytotoxic effector functions. CD8+ cytotoxic T cells play an important role in inducing cell death of tumor cells, as well as cells infected by virus, bacteria or parasites.
