General description
Protein kinase C zeta type (EC 2.7.11.13; UniProt Q05513; also known as nPKC-zeta, PKC-zeta) is encoded by the PRKCZ (also known as PKC2) gene (Gene ID 5590) in human. Protein kinase Cs (PKCs) are threonine/serine kinases that play important roles in cellular signal transductions. PKC isozymes are classified into three major classes based on their domain compositions and activator requirements. The conventional PKCs (cPKCs) include the alpha, beta I, beta II, and gamma isoforms. The novel PKCs (nPKCs) include the delta, epsilon, eta, and theta isoforms. Atypical PKCs (aPKCs) include the lambda (mouse ortholog to human PKC-lambda is called PKC-iota) and zeta isoforms. Both cPKCs and nPKCs bind diacylglycerol (DAG) via their C1 domain, while only cPKCs, but not nPKCs bind Ca+2 via their C2 domain. The aPKCs do not bind either DAG or calcium for activity. All three classes of PKCs contain a pseudosubstrate sequence in their N-terminal region that keeps the substrate-binding cavity occupied via intramolecular interaction when the enzymes are in their inactive conformation. Conformation change upon enzyme activation frees the substrate-binding site.
