Biochem/physiol Actions
Human natural killer-1 (HNK-1)/cluster of differentiation (CD57) is also found on some B-cell lines and B lymphocytic leukemia (B-CLL) melanoma lines. The antigen is also detected in lymphoid tissues, certain neuronal subsets, peripheral nerve Schwann cells, and normal, benign, and malignant prostate. Neurofibromas, schwannomas, traumatic neuromassmall cell lung carcinomas frequently express HNK-1. CD57/HNK-1 appears to be expressed in many species ranging from insects to humans. It is thought to be involved in cellular adhesion events and is expressed on several adhesion related proteins including most neural immunoglobulins superfamily molecules.

General description
Mouse Monoclonal Anti-Human CD57/HNK-1 (mouse IgM isotype) is derived from the VC1.1 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a homogenate of cat cerebral cortical area 17. Human natural killer-1 (HNK-1) also called β-1,3-glucuronyltransferase (B3GAT1), is mapped to human chromosome 11q25 and belongs to the glucuronyltransferase gene family. CD57/ HNK-1 is expressed on a subpopulation (15-20%) of peripheral blood mononuclear cells. Expression of the antigen was described for about 60% of NK active cells and for a T-cell subset.

Specificity
Recognizes the CD57/HNK-1 human myeloid cell associated surface glycoprotein. The epitope recognized is an N-linked carbohydrate which is present in a variety of glycoproteins and in some glycolipids. It is resistant to formalin fixation and paraffin embedding. VC1.1 antibody and the HNK-1 (Leu7) antibody inhibit the binding of each other. The antibody recognizes myelin associated glycoprotein in some species and a high molecular weight chondroitin sulphate proteoglycan. 5th Workshop: code no. NK 67
Target description
CD57/HNK-1 is expressed on a subpopulation of 15-20% of peripheral blood mononuclear cells, about 60% of NK active cells and on a subset of T cells.
