General description
Cell division control protein 45 homolog (UniProt O75419; also known as PORC-PI-1) is encoded by the CDC45 (also known as CDC45L, CDC45L2, UNQ374/PRO710) gene (Gene ID 8318) in human. Recruitment of replication initiation proteins at the G1/S transition enables origin DNA unwinding and loading of DNA polymerases. CDC45 is a replication initiation protein that plays an essential role in the establishment and progression of the replication fork. Cdc45 associates with the heterohexameric Mcm2-7 helicase and the GINS complex (go-ichi-nii-san, Japanese for 5-1-2-3, for the subunits Sld5, Psf1, Psf2 and Pfs3) to form the CMG (Cdc45-Mcm2-7-GINS) complex. CMG represents the eukaryotic replicative DNA helicase that unwinds origins of replication and drives the replication fork. CDC45 is also reported to interact with several other replication proteins, including DNA helicase B (DHB), Mcm10, Orc2, TopPB1, and DNA polymerases alpha, delta and epsilon.
