General description
Myristoylated Alanine-Rich C-Kinase Substrate (MARCKS) is a widely expressed substrate of PKC. MARCKS is essential in neuronal development, as mice deficient in it die shortly after birth. Through its basic effector region, MARCKS associates with acidic membrane phospholipids, actin filaments, and calmodulin. Poly (ADP-ribose) also binds the basic effector region, blocking calmodulin binding, membrane association, and phosphorylation by PKC, suggesting that MARCKS may be an effector of the DNA-damage signal transduced by poly (ADP-ribose).
Immunogen
Synthetic peptide corresponding to amino acids surrounding the phosphoSer152/156 of rat MARCKS.
Epitope: phosphoSer 152/156
Linkage
Replaces: AB5879
Physical form
Affinity purified
Purified rabbit polyclonal IgG in 10 mM Hepes, pH 7.5, with 150 mM NaCl, 100 µg/mL BSA and 50% glycerol.
Quality
Routinely evaluated by Western Blot on EGF treated A431 lysates.
Western Blot Analysis: 1:500 dilution of this lot detected phosphorylated MARCKS on 10 µg of EGF treated A431 lysates.
Specificity
MARCKS, phosphoSer152/156.
Storage and Stability
Stable at -20°C in undiluted for up to 6 months from date of receipt. Avoid repeated freeze/thaw cycles. Do not store in a self-defrosting freezer.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
