Application

Detect phospho-Bcl-2 (Ser70) using this Anti-phospho-Bcl-2 Antibody, clone 69-10C-2-10C-18 validated for use in WB, IP, FC.

Immunoprecipitation Analysis: 1ug from a previous lot immunoprecipitated phosphorylated Bcl-2 from 100 ug of jurkat lysate treated with paciltaxel. Arrow indicates phospho-Bcl-2 (~26kD).

Flow Cytometry Analysis: 0.1ug/ml from a previous lot was used to detect Bcl-2 in Jurkat cell lysate.

General description

The Bcl name is derived from B-cell chronic lymphocytic leukemia, and was given to genes involved in that disease. Members of the Bcl-2 family share homologous domains (BH) through which they are able to homo- and hetero-dimerize. Bcl-2 plays an important role in cytochrome c release from the mitochondria; it also binds Apaf1, inhibiting Apaf-1 association with caspase-9. Bcl-2 overexpression in indicated in a number of human cancers, including melanoma, breast, prostate, and lung, and it correlates with resistance to cancer treatment (presumably due to decreased apoptosis). Bcl-2 has also been implicated in schizophrenia and autoimmune disorders. Bcl-2 phosphorylated at Ser-70 plays a critial role in the translocation of Bax from the cytosol to the mitochondria, and this may regulate NO-induced neuronal apoptosis. The molecular mechanism of Bcl-2 phosphorylation and its relationship to Bax is largely unknown.

Evaluated by Western Blot in paclitaxel treated and untreated Jurkat cell lysate.

Western Blot Analysis: 1:1,000 dilution of this antibody detected Bcl-2 on 10 µg of Jurkat cell lysate.

Specificity

This antibody recognizes phosphorylated Bcl-2 at Ser70.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
