General description

Polycomb complex protein BMI-1 (UniProt P25916; also known as B lymphoma Mo-MLV insertion region 1, Polycomb group RING finger protein 4) is encoded by the Bmi1 (also known as AW546694, Bmi-1, Pcgf4) gene (Gene ID 12151) in murine species. Bmi1 is a member of the polycomb group (PcG) family of evolutionarily conserved gene silencers involved in cell fate decisions and body pattern during development. Bmi1 is an important regulator of self-renewal of hematopoietic and neural stem cells, as well as cancer stem cells (CSCs). Bmi1 is a main constituent of the polycomb repressive complex 1 (PRC1) and pariticipates in PRC-mediated gene silencing by promoting monoubiquitination of H2A. Bmi1 overexpression is reported in many different human cancers and high level of Bmi1 is considered as a poor prognostic marker. Bmi1 targets genes involved in transforming growth factor-β/bone morphogenetic protein (TGF-beta/BMP), endoplasmic reticulum (ER) stress, and WNT pathways. Bmi itself is transcriptionally regulated by c-Myc and E2F1 transcription factors.
