Application
Transfection Reagent I has been used in the preparation of liposomes along with cholesterol for fluorescence-based live-cell imaging studies in the mouse embryonic fibroblasts cells. It may be used for the transfection of DNAzyme (Dz13) into veins.
Biochem/physiol Actions
Transfection Reagent I is used in large unilamellar cationic vesicles (LUVs) preparation and lipoplex generation. It improves transfection efficiency. DOTAP/DOPE (1,2-dioleoyloxypropyl)-N,N,N-trimethylammonium chloride /(1,2-dioleoyl-sn-glycero-3-phosphoethanolamine) is safe transfection agent for human administration. It inhibits the blood vessel thickening and is a promising candidate for use in coronary artery bypass grafting (CABG).
General description
Transfection Reagent I comprises the cationic lipid 1,2-dioleoyloxypropyl)-N,N,N-trimethylammonium chloride (DOTAP) and the neutral lipid DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine. DOTAP is a surfactant with double chain quaternary ammonium. The DOTAP/DOPE liposome takes lamellar and inverted hexagonal structures.
