Target description

Retinoic acid early transcript 1E (UniProt: Q8TD07; also known as Lymphocyte effector toxicity activation ligand, NKG2D ligand 4, N2DL-4, NKG2DL4, RAE-1-like transcript 4, UL16-binding protein 4, ULBP-4) is encoded by the RAET1E (also known as LETAL, N2DL4, ULBP4, UNQ1867/PRO4303) gene (Gene ID: 135250) in human. ULBP-4 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-30), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 31-225), a transmembrane domain (aa 226-248), and a cytoplasmic domain (aa 249-263). It is predominantly expressed in the skin, but also expressed in testis and trachea. Its levels are upregulated in tumor cells of different origins and its expression progressively decreased after treatment of tumor cells with retinoic acid. ULBP-4 also serves as a functional ligand for NKG2D. ULBP-4 is reported to bind and co-stimulate NKG2D-expressing effector cells including NK cells, NKT cells, CD8+ T cells. ULBP-4 has a MHC class I alpha-1 like (aa 31-116) and MHC class I alpha-2 like (117-207) regions that down-regulate the cell surface expression of Killer Cell Lectin Like Receptor K1 (KLRK1). Six different isoforms of ULBP-4 have been described that are produced by alternative splicing.
