General description

CREB-regulated transcription coactivator 2 (CRTC2) is a transcriptional coactivator for CREB1 which is a cAMP (CRE) site responder. CRTC2 is also known as TORC-2 (transducer of regulated cAMP response element-binding protein 2) and CRTC2 regulates gluconeogenesis as well as steroidogenic processes, and is also a coactivator for mitochondrial biogenesis in muscle cells. Localization is cytoplasmic but relocates to the nucleus in response to cAMP and glucagon levels. CRTC2 is highly expressed in thymus, spleen, muscle, and lung; it is also expressed in various cancers as well as insulinomas. Low levels of expression are found in the brain, colon, heart and kidney, with the lowest levels being in the liver and pancreas. Activity is regulated via phosphorylation. CRTC2 is inactive when phosphorylated (meaning that is does not bind CREB and assist in gene transcription) and active when dephosphorylated. EMD-Millipore's Anti-CRTC2 mouse monoclonal, clone 5B10, has been tested in western blots against human HEK292 and HeLa cell extracts, in immunohistochemistry in paraffin embedded human ovary and lung cancer tissue, and in immunofluorescent immunocytochemistry and flow cytometry on HeLa cells in culture.

Immunogen

Purified protein fragment of human CRTC2 expressed in E. coli.

Physical form

Mouse monoclonal IgG2b ascitic fluid containing up to 0.1% sodium azide.

Unpurified

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CRTC2 in 10 µg of HeLa cell lysate.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
