General description
Kallikrein-5/KLK5, or also known as Kallikrein-related peptidase 5, Kallikrein-like protein 2, KLK-L2, and formerly known as Stratum corneum tryptic enzyme (SCTE), is an important enzyme encoded by the gene KLK5/KLK-L2/SCTE. KLK5 is a secreted serine protease enzyme that is expressed in many tissues including the skin, breast and brain. In the skin, Kallikreins-related peptidases (KLKs) are proteases that are important for the desquamation process of the skin (cell shedding). KLK5 is considered as one of the most important KLKs in skin desquamation because KLK5 also regulates the activation of other Kallikreins needed for cell shedding. In disease, KLK5 displays aberrant expression in many cancers. However, expression of KLK5 in some cancer cells appears to suppress tumor growth and activity via reduced RhoA activity and the modulation the mevalonate pathway. This is particularly true in breast cancer and efforts are examining methods to increase KLK5 expression in breast cancer as a form of treatment.
