Application
Polysaccharide conjugation: CDAP Ready Made Solution (100mg/mL in acetonitrile) should be added in a ratio of 1mg CDAP to 1mg polysaccharide while vortexing (Example: per 1mg polysaccharide add 10μL of CDAP Ready Made Solution). Add 0.2M triethylamine, 10μL per 1mg polysaccharide to raise the pH of the reaction.
Biochem/physiol Actions
CDAP is a cyanylation reagent for protein sulfhydryl groups which is used to prepare protein-polysaccharide conjugates. CDAP is also an activating agent for polysaccharide resins and is used for the conjugation of lipopolysaccharides while retaining their endotoxic activity.CDAP is considered to be a less toxic reagent as compared to cyanogen bromide (CNBr) (a known polysaccharides activator). In addition, CDAP is easier to use as it can be employed at a lower pH and has fewer side reactions. It is known that CDAP polysaccharide activation efficiency is optimal at pH 9-10. It was also reported that direct conjugation of protein to CDAP-activated polysaccharides can be performed under mildly alkaline conditions (pH 7-9). It has also been reported that proteins could also be conjugated to CDAP-activated polysaccharides at pH 5.
