β-Lactamasehydrolyzesβ-lactumantibioticsandisthechief cause ofresistance to β-lactam antibiotics development by bacteria. Mutationsin theβ-lactamases are associated with extended-spectrum β-lactamases (ESBLs).
General description
β-Lactamaseproduced bybacteriashares sequence homology topenicillin-binding proteins. This enzyme is found inGram-negativebacteria. β-Lactamaseshave four molecularclasses, named A, B, C, and D. A, C, and D form an acyl-enzyme viaactive site serine residue.Class B β-lactamasesaremetalloenzymes, which have a zinc ion attheiractive site for β-lactam hydrolysis.
