General description

Forkhead box H1 (FOXH1) is a member of the HNF–3FKH family of transcriptional activators. FOXH1 is a winged helix with two loops on the C-terminal side expressed in all human tissue. FOXH1 is an embryonic transcription regulator and is an essential transcriptional co-activator that binds with SMAD2 to activate activin, and therefore induce the gooseciod (GSC) promoter. After FOXH1 binds to SMAD4, activin response element is activated and TGF-β is stimulated. FOXH1 also interacts with NKX2-5 to form an essential component of anterior heart field development. FOXH1 is associated with complex brain malfunctions due to incomplete cleavage of the prosencephalon, and in developmental delays resulting in facial abnormalities such as cyclopia and severe cleft lip or palate.

Purified rabbit polyclonal antibody precipitated in a solution of 50% saturated ammonium sulfate and PBS containing no preservatives.

Evaluated by immunocytochemistry on NIH/3T3, HeLa, and A431 cells.

Immunocytochemistry:
1:500 dilution of this antibody was used to detect FOXH1 in NIH/3T3, HeLa and A431 cells.

Specificity

This antibody recognizes FOXH1.

Storage and Stability

Stable for 1 year at 2-8°C from date of receipt.
To reconstitute the antibody, centrifuge the antibody at a moderate speed (5,000 rpm) for 5 minutes. Carefully remove the ammonium sulfate/PBS buffer solution and discard; 10µL of residual ammonium sulfate solution will not affect the re-suspension of the antibody. Do not let the protein pellet dry, as severe loss of antibody reactivity can occur. Re-suspend the antibody pellet in 100L either standard PBS or TBS (pH 7.3-7.5). DO NOT VORTEX. Mix by gentle stirring with a wide pipet tip or gentle finger-tapping. Let the precipitated antibody rehydrate for 1 hour at 4-25°C prior to use. Small particles of precipitated antibody that fail to re-suspend are normal. Vials are overfilled to compensate for any losses.
