ONO-2506 (arundic acid; (R)-2-propyloctanoic acid) is an inhibitory astrocytes-modulating agent that exerts glioprotective effects against amyloid-β-peptide (Aβ)-induced glial death in astrocyte cultures (50 µM ONO-2506 against 200 µM Aβ25-35, 24 hr; 1321N1 astrocytes) by decreasing both the intracellular and extracellular S100B levels. Blockade of astrocytic activation by ONO-2506 in vivo is shown to offer therapeutic efficacy in animal models of normal tension glaucoma (10 mg/kg po. mice), spinal cord injury (20 mg/kg/d ip or iv rats), brain seizure, ischemic stroke (10 mg/kg/d ip or iv rats), and Parkinson′s disease (30 mg/kg ip mice).
