Detect Phosphacan using this Anti-Phosphacan Antibody validated for use in ELISA, WB, IH(P).

Samples must be digested with chondroitinase prior to running on SDS gels because undigested phosphacan is too large for most gels. Treatment is at a concentration of chondroitinase of 10U/mL in Tris-HCL pH 8.0. Make tissue or cell extract in 20-50mM Tris pH 7.6-8.0 with 0.15M NaCl in the presence of protease inhibitors. Add 1 microliter of enzyme to 30 microliters of extract and incubate 30 minutes at 37C. Then add SDS sample buffer, heat or boil sample as normal for SDS reducing samples.
Immunohistochemistry: 1:20 Works on cold methanol or paraformaldehyde fixed tissues. Works on paraffin embedded tissues. Chondrotinase treatment of tissue consists of treating mounted tissue section with a solution of PBS plus 0.5U/mL of Chondroitinase ABC for 30 minutes at room temperature prior to blocking and antibody staining.
Immunocytochemistry: not tested
ELISA: Excellent for core and monomer protein.
Optimal working dilutions must be determined by the end user.

General description
Phosphacan is a soluble nervous tissue-specific proteoglycan that is thought to regulate axonal outgrowth. It is synthesized by glia and binds to neurons and to the neural cell adhesion molecules tenascin, N-CAM or NG-CAM but not to laminin and fibronectin. Phosphacan acts as a potent inhibitor of cell adhesion and neurite outgrowth.

Mixture of chicken chondroitin sulfate proteoglycans.
