NovaTaq DNA polymerase has been used to check the success of the ligation step during cloning. It has also been used in the PCR amplification of cDNA.NovaTaq DNA Polymerase can be used in standard polymerase chain reaction (PCR) amplification protocols. It is also suitable for colony PCR for screening bacterial colonies.

General description
NovaTaq DNA polymerase is a recombinant form of Thermus aquaticus DNA polymerase. This enzyme is a non-proofreading DNA polymerase. NovaTaq DNA polymerase exhibits 5′-3′ DNA polymerase activity and lacks 3′- 5′ exonuclease activity. The preparation is >95% homogenous by sodium dodecyl sulfate-poly acrylamide gel electrophoresis (SDS-PAGE) and lacks detectable endonuclease activities. It is tested for several quality control assays. This enzyme produces PCR products with 3′-dA overhangs, suitable for cloning with Novagen®Perfectly Blunt®, AccepTor, and LIC Vector Kits. Unit Definition
One unit is defined as the amount of enzyme that will catalyze the incorporation of 10 nmol of dNTP into acid-insoluble form in 30 min at 74°C, in a reaction containing 25 mM TAPS (tris-[hydroxymethyl]-methyl-amino-propane-sulfonic acid, sodium salt), pH 9.3 at 25°C, 50 mM KCl, 2 mM MgCl₂, 1 mM 2-mercaptoethanol, 0.2 mM dATP, dGTP, and dTTP, 0.1 µM [A-32P]dCTP, and activated salmon sperm DNA.
