Western Blotting Analysis: A 1:500 dilution from a representative lot detected TMIGD1 in 10 µg of human kidney tissue lysate and Raji membrane extract.
Western Blotting Analysis: A representative lot detected the overexpression of a c-myc tagged murine TMIDG1 in transfected HEK-293 cells, as well as the endogenous TMIDG1 in mock-transfected HEK-293 cells. General description

Transmembrane and immunoglobulin domain-containing protein 1 (UniProt Q6UXZ0) is encoded by the TMIGD1 (also known as TMIGD) gene (ORF UNQ9372/PRO34164; Gene ID 388364) in human. TMIGD1 protein is an adhesion molecule expressed in renal tubular epithelial cells and is highly conserved between human and murine species. TMIGD1 regulates renal epithelial cell trans-epithelial electric resistance, permeability, migration and cell morphology, and protects renal epithelial cells from oxidative stress and nutrient deprivation-induced injury. Hydrogen peroxide is shown to induce TMIGD1 ubiquitination and degradation. In addition, TMIGD1 expression is downregulated in mouse acute kidney injury (AKI) and in deoxy-corticosterone acetate (DOCA) and sodium chloride (DOCA-salt)-induced chronic hypertensive kidney disease models. TMIGD1 is produced with a signal peptide sequence (a.a. 1-29), the removal of which yields the mature single-transmembrane (a.a. 221-241) protein with a large extracellular region (a.a. 30-220) and a short cytoplasmic tail (a.a. 242-262). The extracellular region contains two C2-type Ig-like domains (a.a. 30-114 & 122-207) that mediate TMIGD1 dimerization.

This polyclonal antiserum detects glycosylated as well as unglycosylated and deglycosylated TMIGD1.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Target description

~30/39/52 kDa observed. Target bands appear larger than the calculated molecular weights of 29.19/25.82 kDa (pro-/mature form of isoform 1) and 24.28/20.91 kDa (pro-/mature form of isoform 2) due to glycosylation. Uncharacterized band(s) may appear in some lysates.
