General description
PSD-95/SAP90-binding protein 1 (SAPAP1) is a member of a protein family whose members specifically interact with PSD-95/SAP90, a membrane-associated guanylate kinase localized at postsynaptic density (PSD) in neuronal cells. Like the other SAPAP proteins, SAPAP1 is thought to be an adaptor protein that also interacts with different synaptic scaffolding proteins, cytoskeletal and signaling components, such as focal adhesion kinase (FAK) and proline-rich tyrosine kinase 2 (PYK2). SAPAP1 mRNA is targeted to cell bodies in a similar manner to SAPAP2 and -4, whereas SAPAP3 mRNA is detected mainly in cell bodies. SAPAP1 protein however, is targeted to the synapse and is not reliant on the synaptic localization of PSD-95 or the synaptic scaffolding molecule (S-SCAM).
Immunogen
KLH-conjugated linear peptide corresponding to human SAPAP1.
Other Notes
Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
Physical form
Purified rabbit polyclonal in buffer containing PBS with 0.02% sodium azide.
Antigen Affinity Purified
Quality
Evaluated by Western Blot in human brain tissue lysate.
Western Blot Analysis: 1 µg/mL of this antibody detected SAPAP1 in 10 µg of human brain tissue lysate.
Specificity
At least three isoforms of SAPAP1 are known to exist. This SAPAP1 antibody will not cross-react with other SAPAP proteins.
Storage and Stability
Stable for 1 year at 2-8°C from date of receipt.
Target description
~95 kDa observed. Uniprot describes 7 isoforms ranging between ~70 and ~109 kDa An uncharacterized band may appear at ~58 kDa in some lysates. The calculated molecular weight is 109 kDa, however SAPAP1 has been shown as a ~95 kDa band in western blots. (Naisbitt, S., et al. (1997). 17(15):5687-5696.)
