General description
The protein called Peroxiredoxin-2, or in older literature Thiol-specific antioxidant protein, TSA, Thioredoxin peroxidase 1, Thioredoxin-dependent peroxide reductase 1, Natural killer cell-enhancing factor B, PRP, or NKEF-B and encoded by the human gene PRDX2/NKEFB/TDPX1 plays a role in controlling the oxidation state of the cell by reducing peroxides via the Thioredoxin system. Peroxides are produced during normal metabolism and if not reduced, their built up can lead to oxidative stress and cell death. Interestingly Peroxirdoxin-2 can be inactivated itself by oxidative stress on Cys-51 thus its levels are tightly controlled. Peroxiredoxin-2 is particularly important in neurons where it acts as an anti-apoptotic factor but the protein is widely expressed and localizes to the cytoplasm in most cells. Because Peroxiredoxin-2 mediates H2O2 signaling it is considered an anti-aging and anti-cancer protein, but surprisingly recent research in yeast indicates that Peroxiredoxin-2 potentiates H202 levels and signaling by specifically inhibiting other Thioredoxin enzymes via their oxidation. Thus Peroxiredoxin-2 can both inhibit and promote peroxide metabolism depending upon the cellular microenvironment and cell systems being studied.
