General description
The protein named Microcephalin and encoded by the human gene named MCPH1 is a DNA/chromatin binding protein implicated in chromosome condensation and DNA damage induced cellular responses. MCPH1 plays a role in neurogenesis and is involved in the regulation of the size of the cerebral cortex. MCPH1 interacts with several proteins including H2AFX under DNA damage conditions and CDC27 during normal cell growth. MCPH1 is expressed cytoplasmically in a variety of cell types and tissues and is particularly expressed in the fetal brain, liver and kidney. Defects in MCPH1 can give rise to microcephaly a disease where the head circumference is markedly reduced and effected patients are mentally retarded, showing a high number of prophase-like cells in cytological examinations. In addition, cell divisions are irregular and demonstrate a delay in post-mitotic chromosome decondensation. Interestingly, recent research demonstrates that MCPH1 also functions as a tumor suppressor that is regulated by the microRNA miR-27a.
