General description

Arpin (UniProt Q7Z6K5; also known as Arp2/3 inhibition protein, UPF0552 protein C15orf38) is encoded by the ARPIN (also known as C15orf38) gene (Gene ID 348110) in human. Cell migration requires the generation of branched actin networks that power the protrusion of the plasma membrane in lamellipodia. The actin-related proteins 2 and 3 (Arp2/3) complex nucleates actin filaments by binding to the side of an existing filament and initiating branch formation. Arpin functions as a negative regulator of Arp2/3 activity, allowing cells to control directional persistence of migration by fine-tuning actin nucleation activity at the leading edge of the lamellipodium. Arpin depletion in both mammalian cells and Dictyostelium discoideum amoeba is reported to result in straighter trajectories. Arpin microinjection is demonstrated to induce fish keratocytes to turn.
