Immunofluorescence Analysis: A representative lot immunostained predominantly renal collecting ducts by fluorescent immunohistochemistry using kidney sections from postnatal P12 Pkd1v/v ADPKD mice (Kwon, M., et al. (2012). Proc. Natl. Acad. Sci. U. S. A. 109(52):21462-21467).
Immunohistochemistry Analysis: A representative lot detected a time-dependent induction of AGS3/GPSM1 immunoreactivity co-localized with that of the proliferation marker Ki-67 in formalin-fixed, paraffin-embedded kidney sections from rats following ischemia-reperfusion injury (IRI) initiated by a 30-min occlusion of renal blood flow via bilateral renal artery clamping (Regner, K.R., et al. (2011). FASEB J. 25(6):1844-1855).
Western Blotting Analysis: A representative lot detected developmental stage-dependent renal AGS3/GPSM1 expression level. High level was seen in E13.5 embryonic kidney and greatly reduced among postnatal mice with low-to-undetectable levels around postnatal day P28. Postnatal renal AGS3/GPSM1 levels were found elevated and remained detectable up to P28 among Pkd1v/v autosomal dominant polycystic kidney disease (ADPKD) mice expressing noncleavable polycystin-1/PC1 (Kwon, M., et al. (2012). Proc. Natl. Acad. Sci. U. S. A. 109(52):21462-21467).

General description

G-protein-signaling modulator 1 (UniProt Q9R080; also known as Activator of G-protein signaling 3) is encoded by the Gpsm1 (also known as Ags3) gene (Gene ID 246254) in rat species. AGS3/GPSM1 is an evolutionarily conserved protein with four G-protein regulatory (GPR) GoLoco motifs (a.a. 493-515, 546-568, 594-616, 628-650 of murine GPSM1) and functions as a guanine nucleotide dissociation inhibitor (GDI). GPSM1 increases heteromeric polycystin-1/polycystin-2 ion channel activity via Gβγ subunits. GPSM1 is involved in regulating mitotic spindle orientation, cell polarity, and adenylyl cyclase activity. GPSM1 plays a role in renal epithelial cell repair following renal injury and GPSM1 induction in polycystic kidney disease (PKD) and may be a critical modulator of the renal cystogenic process.

Western Blotting Analysis: A 1:500 dilution of this antibody detected AGS3/GPSM1 in 10 µg of E13-14 embryonic mouse brain tissue lysate.

Specificity

This polyclonal antiserum immunostained predominantly renal collecting ducts of postnatal P12 ADPKD mice. Immunogen peptide blocking eliminated antibody stianing (Kwon, M., et al. (2012). Proc. Natl. Acad. Sci. U. S. A. 109(52):21462-21467). This antiserum targets a STK11/LKB1-interacting region sequence present in rat AGS3/GPSM1 isoform 1 and isoform 4 (isoform Long), 28% present in isoform 3 (Short-2), but completely absent from isoforms 2 (Short-1) reported by UniProt (Q9R080). Target sequence is also present in mouse isoforms 1, 3, 4, and 5, but absent from isoform 2 ( (UniProt Q6IR34).

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
