General description
Arf-GAP with dual PH domain-containing protein 1 (ADAP1), also known as p42(IP4), PIP3BP, or centaurin-alpha1, is a highly abundant brain protein that has been detected in the hippocampus, cerebellum, cortex, and hypothalamus. It is an actin-binding protein that has been localized to the nucleus, cytoplasm, plasma membrane, and mitochondria. ADAP1 contains a zinc finger motif and two pleckstrin homology domains. It functions as a receptor for inositol(1,3,4,5)tetrakisphosphate and phosphatidylinositol(3,4,5)trisphosphate second messengers, and also binds to and becomes phosphorylated by all isoforms of PKC. It has been linked to calcium transport in the mitochondria and RNA metabolism in the nucleus. In addition, it has been shown that ADAP1 modulates the development and differentiation of dendrites in hippocampal cells, as it is involved in the formation of dendritic filopodia and spine-like protrusions. It has also been reported that ADAP1 is upregulated in Alzheimer’s disease.
