Analysis Note

ControlNIH/3T3 cell extract
Included Positive Antigen Control: Catalog # 12-305, 3T3/A31 Cell Lysate. Add 2.5 µL of 2-mercaptoethanol per 100 µL of lysate and boil for 5 minutes to reduce the preparation. Load 20 µg of reduced lysate per lane for minigels.

Application

Research Sub CategoryHistones

Use Anti-HDAC1 Antibody (Rabbit Polyclonal Antibody) validated in EA, ICC, IP, WB to detect HDAC1 also known as histone deacetylase 1.

Research CategoryEpigenetics & Nuclear Function

Immunoprecipitation:
5 µg of a previous lot immunoprecipitated HDAC1 from 500 µg of 3T3/A31 RIPA lysate.

Immunocytochemistry:
This antibody has been reported to immunostain HDAC1 in 1% paraformaldehyde fixed, 0.1% Triton X-100 permeabilized 3T3 cells.

HDAC Activity:
This antibody has been reported to immunoprecipitate active HDAC.

General description

Mammalian HADC1 (also designated HD1), HDAC2 (also designated mammalian RPD3) and HDAC3, all are related to the yeast transcriptional regulator Rpd3p, are identified as histone deacetylases. The deacetylation of histones is associated with transcriptional silencing.

Immunogen

6X-His-tagged fragment representing amino acids 53-482 of mouse histone deacetylase 1 (HDAC1).

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.

Physical form

Protein A chromatography

Purified rabbit polyclonal IgG in buffer containing 0.1 M Tris-glycine, pH 7.4, 0.15 M NaCl, containing 0.05% sodium azide. Frozen at -20°C.

Format: Purified

Quality

Routinely evaluated by western blot on RIPA lysates from 3T3/A31 cells.

Western Blot Analysis:
0.5-2 µg/mL of this lot detected HDAC1 in RIPA lysates from 3T3/A31 cells.

Specificity

Recognizes and is specific for HDAC1, Mr 65 kDa. May detects a non-specific protein at ~35 kDa.

Oher species cross-reactivity not tested.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.

Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
