General description

Plakophilin-3 (UniProt: Q9Y446; also known as PKP3) is encoded by the PKP3 gene (Gene ID: 11187) in human. Plakophilin 3 a novel armadillo-like protein present in nuclei and desmosomes of epithelial cells and dendritic reticular cells of lymphatic follicles. Plakophilin-3 is an Armadillo-like protein whose expression pattern is largely restricted to epithelial cell types and plays a role in junctional plaques. It is localized in the desmosomal plaques and in the nucleus. Plakophilin-3 contains a central Arm repeat domain, containing 10 Arm repeats and a short C-terminal domain. Plakophilin proteins act in a cellular desmosome-dependent adhesion and signaling pathways. It is shown to bind directly to the desmosomal cadherins Dsg1, Dsg2, Dsg3, Dsc3a, and Dsc3b, and possibly also to Dsc1a and Dsc2a. Two isoforms of Plakophiln-3 have been described that are produced by alternative splicing. Isoform PKP3a is found in desmosomes of most simple and stratified epithelia and PKP3b is abundant in the stratified epithelial cells, but not in simple epithelial cells.
